I have a doubt, when i configure the moh in asterisk, i set a directory in a moh context in musiconhold.conf. But if i have some wav files in de directory, which and how of all chooses asterisk to play?

The format doesn’t matter as Asterisk can play the file. In case there is a problem, the a warning is shown in the cli, I think (at least if called explicitly with playback).
Asterisk will play the format that it thinks least constly to transcode to tthe current channel.
